Abstract:
Background/Aims: Autophagy is a dynamic catabolic process that maintains celluarhomeostasis. Whether it plays a role in promoting cell survival or cell deathin the process of renal ischemia/reperfusion(I/R) remains controversial, partlybecause renal autophagy is usually examined at a certain time point. Therefore,monitoring of the whole time course of autophagy and apoptosis may help betterunderstand the role of autophagy in renal I/R.

背景/目的: 自噬是维持细胞稳态的动态分解代谢过程。 它是否在肾缺血/再灌注(I / R)过程中在促进细胞存活或细胞死亡中起作用仍有争议,部分是因为通常在某一时间点检查肾自噬。因此,监测自噬和凋亡的整个时间过程可能有助于更好地了解自噬在肾I / R中的作用。

Methods: Autophagy and apoptosis were detected after mice were subjected tobilateral renal ischemia followed by 0-h to 7-day reperfusion, exposure ofTCMK-1 cells to 24-h hypoxia, and 2 to 24-h reoxygenation. The effect ofautophagy on apoptosis was assessed in the presence of autophagy inhibitor3-methyladenine3-MAand autophagy activator rapamycin.
方法: 老鼠接受双侧肾缺血处理,之后0h到7d再灌注,TCMK-1细胞接受24小时缺氧暴露,2到24小时再氧化,然后检测自噬和凋亡。在自体吞噬抑制剂3-甲基腺嘌呤(3-MA)和自噬激活剂雷帕霉素的存在下评估自噬对细胞凋亡的影响。

Results:Earlier than apoptosis autophagy increased from 2-hreperfusion,reached the maximum at day 2, and then began declining from day 3when renal damage had nearly recovered to normal. Exposure to 24-h Hypoxiainduced autophagy markedly, but it decreased drastically after 4 and 8 –hreoxygenation(H/R) in vitro.  Incontrast, activation of autophagy by rapamycin significantly alleviated renaltissue damage and tubular cell apoptosis in the two models.

Conclusion: Autophagy was induced in a time-dependent manner and occurredearlier than the onset of cell apoptosis as an early response that played arenoprotective role during renal I/R and cell H/R. Up-regulation  of autophagy may prove to be a potential strategy for the treatment ofacute kidney injury.

结论: 自噬作用以时间依赖性方式诱导并且发生早于细胞凋亡的发作,作为在肾I / R和细胞H / R期间发挥肾脏保护作用的早期应答。自噬的上调可能证明是治疗急性肾损伤的潜在策略。
